چکیده

Tang, Tong, Mei Hua Gao, David M. Roth, Tracy Guo, and H. Kirk Hammond. Adenylyl cyclase type VI corrects cardiac sarcoplasmic reticulum calcium uptake defects in cardiomyopathy. Am J Physiol Heart Circ Physiol 287: H1906–H1912, 2004. First published July 8, 2004; doi:10.1152/ajpheart.00356.2004.—Calcium malfunction plays a central role in heart failure. Here, we provide evidence that adenylyl cyclase type VI restores sarco(endo)plasmic reticulum 2a (SERCA2a) affinity for calcium and maximum velocity of cardiac calcium uptake by sarcoplasmic reticulum in murine dilated cardiomyopathy. Restoration of normal SERCA2a affinity for calcium is associated not only with decreased phospholamban protein expression but also with increased phospholamban phosphorylation by PKA activation. The ratio of phosphorylated ryanodine receptor 2 (RyR2) to RyR2 protein was increased, but the amount of phosphorylated RyR2 was unaffected. These data provide a possible mechanism by which adenylyl cyclase type VI (in contrast to other signaling elements associated with increased cAMP generation) has a salutary effect in the failing heart.

Increased cardiac adenylyl cyclase expression is associated with increased survival after myocardial infarction.

BACKGROUND Cardiac-directed expression of adenylyl cyclase type VI (AC(VI)) in mice results in structurally normal hearts with normal basal heart rate and function but increased responses to catecholamine stimulation. We tested the hypothesis that increased left ventricular (LV) AC(VI) content would increase mortality after acute myocardial infarction (MI). METHODS AND RESULTS Transgenic mice...

The relation between calcium ion transport and adenylate cyclase activity in myocardial sarcoplasmic-reticulum preparations.

The regulation of CaQ2 concentration in relation to myofibrils plays a central role in the production of contraction and relaxation in cardiac muscle (Langer, 1968). The calcium pump of the sarcoplasmic reticulum is a major factor producing relaxation by removing Ca2+ from the region of the myofibrils, and may secondarily produce changes in Ca2+ release on activation leading to alterations in m...

Alterations in cardiac contractile performance and sarcoplasmic reticulum function in sucrose-fed rats is associated with insulin resistance.

Diabetes mellitus (DM) causes the development of a specific cardiomyopathy that results from the metabolic derangements present in DM and manifests as cardiac contractile dysfunction. Although myocardial dysfunction in Type 1 DM has been associated with defects in the function and regulation of the sarcoplasmic reticulum (SR), very little is known about SR function in Type 2 DM. Accordingly, th...

Adenylyl cyclase type 6 deletion decreases left ventricular function via impaired calcium handling.

BACKGROUND Adenylyl cyclases (ACs) are a family of effector molecules for G-protein-coupled receptors. The 2 ACs most abundantly expressed in cardiac myocytes are types 5 (AC5) and 6 (AC6), which have 65% amino acid homology. It has been speculated that coexpression of 2 AC types in cardiac myocytes represents redundancy, but the specific role of AC6 in cardiac physiology and its differences fr...
